Description

Humanin Peptide: Mitochondrial Protection, Neuroprotection, and Metabolic Support

Humanin is a naturally occurring peptide encoded by the mitochondrial 16S ribosomal RNA gene. It consists of 21–24 amino acids, depending on its site of synthesis, and has been widely studied for its potential in cellular protection, metabolic regulation, neuroprotection, and anti-apoptotic activity.

Humanin may bind with intracellular molecules like Bax, actinin-4, and phosphoprotein 8, as well as cell surface G protein-coupled receptors (FPRL-1 and FPRL-2), to protect cells from oxidative stress, apoptosis, and neurodegeneration. Research suggests it may improve mitochondrial function, cognitive performance, insulin sensitivity, and survival under ischemic or hypoxic conditions.

Chemical Makeup

Research and Clinical Insights

Mitochondrial Function & Cellular Protection

  • Humanin may reduce reactive oxygen species (ROS), preserving mitochondrial bioenergetics.

  • Demonstrated cytoprotective effects in retinal pigment epithelial cells under oxidative stress.

Neurological Support & Cognitive Function

  • May inhibit Bax-mediated apoptosis and protect neurons from toxins.

  • Improved learning and memory in murine models with high amyloid levels.

  • Potentially reduces tau protein hyperphosphorylation, relevant in neurodegenerative conditions.

Metabolism & Insulin Sensitivity

  • Shown to enhance insulin sensitivity and glucose tolerance in diabetic and high-fat diet murine models.

  • May reduce weight gain, fasting glucose, and improve energy expenditure.

  • Central administration linked to STAT-3 activation, improving peripheral insulin response.

Ischemia & Hypoxia Protection

  • Protects cells under low oxygen conditions, reducing apoptosis.

  • May reduce ischemic injury and infarct size when administered within critical time windows.

Anxiety & Neuroprotection

  • Binding with FPR2 receptors in the brain may provide anxiolytic effects.

  • Demonstrated potential to protect healthy cells during cancer therapy while targeting malignant cells.
